The vendor is supplying Collaborative Robot Education Training Cart and the FANUC CNC Simulator with Machine Simulation robotic trainers on wheels. Purpose and Objectives: The Department is requesting the purchase of the Rastrum Platform by Inventia Scientific Corporation to advance the understanding of genetic causes and co-morbidities related to connective tissue diseases, including Ehlers-Danlos Syndrome (EDS).
Traditionally, the Department’s research has relied on 2D cell culture methods, which have provided insights into cellular behavior in a flat environment. However, it has become imperative to explore cellular behavior in three-dimensional (3D) contexts to better mimic the in vivo conditions and understand how various extracellular matrices influence phenotypes associated with connective tissue diseases such as hypermobile Ehlers-Danlos Syndrome (hEDS), ultimately leading to the development and testing of therapeutics.
Sole Source Justification: The Rastrum bioprinting platform by Inventia is uniquely suited to meet the Department’s specific needs of their research for several key reasons:
1.High Reproducibility in 3D Matrices: The Rastrum’s proprietary bioprinting technology enables consistent, reproducible creation of 3D matrices. This consistency is crucial for generating reliable data when studying the impact of matrix environments on cell behavior, especially in the context of EDS where the extracellular matrix (ECM) plays a pivotal role in disease manifestation.
2.Customizable Matrices: The ability to tailor matrices, including customizing the density and composition of collagen and other ECM components, is vital for our research. Hypermobile EDS is characterized by alterations in collagen, and the Rastrum allows us to systematically explore how different matrix compositions affect cellular phenotypes in hEDS, which we cannot achieve with conventional 2D culture methods.
3.Development of Novel In Vitro Models: The Rastrum will be instrumental in developing new in vitro models of hypermobile EDS. These models will serve as a complement to our existing in vivo models, offering a platform to more rapidly test hypotheses and generate data while minimizing the confounding variables inherent in animal models.
4.High-Throughput Screening: The ability to perform high-throughput experiments with varying matrix compositions is another unique advantage of the Rastrum system. This high-throughput capability will accelerate our research by allowing for the rapid testing of various ECM conditions in a controlled manner.
5.Integration with Genomic Data: By combining the cellular models developed using the Rastrum with our genomic data, we aim to uncover new insights into the pathology of hypermobile EDS. This integration will enhance our ability to link genetic variants with their downstream effects on cellular behavior in a more physiologically relevant 3D environment
No other manufacturer currently has a system that can meet all the department’s requirements listed above.

In accordance with §11-35-1560 (A) of the SC Consolidated Procurement Code, it is the intent of the Medical University of South Carolina (MUSC) to award without competition, a sole source contract in accordance with Regulation 19-445.2105 (B) (5) to Femtonics Ltd for the purchase of a Femto 3D Atlas Dichroic Acousto-optic Microscope, FemtoSmart Galvo Microscope, Software and accessories.
Purpose and Objectives: The Department is working on a research plan that is centered on building experimentally tested models of cortical circuit development, plasticity, and cognitive function from embryonic development onwards. This entails both simultaneous recording and manipulation of large numbers of cortical neurons, spread out across three-dimensions (3D), during both embryonic development (very low signal-to-noise ratios), as well as throughout the depth of cortex.
Sole Source Justification: To enable simultaneous recordings and manipulations of neurons within a cortical circuit, it is necessary to have a microscope that can: 1) simultaneously record large numbers of neurons across 3D, 2) stably image from neurons in vivo (i.e. accounting for movements) while maintaining high speed, and 3) utilize two different wavelengths of laser light to randomly stimulate or record from each neuron.
All acousto-optic 2p microscopes satisfy (1), as the technology enables rapid (>30 kHz) random-access targeting of a laser beam across 3D space (thereby enabling targeting many neurons, each at >100 Hz). However, most acousto-optic scopes (both custom-made, and from the only other commercial producer) only record a single point at a time. In living tissue, with potential movements, The Department needs to record an area surrounding each neuron to account for movement. Therefore, the speed advantage of the acousto-optic design is reduced. In contrast, the Femto 3D design includes a “drift scanning” mode to enable the rapid imaging of a checkerboard around each neuron. Therefore, this satisfies the requirement of (2), enabling rapid recording of multiple neurons, in living tissue. Finally, the dichroic version of the Femto 3D enables simultaneous stimulation and recording across randomly selected neurons, throughout the depth of cortex. Therefore, this satisfies the requirement for (3). This makes the Femto 3D Atlas the only commercially available microscope that can satisfy all these requirements.
In addition to this specialty microscope, it is also necessary for the Department to have an additional microscope specifically for performing recordings, stably and repeatably. Most current two-photon microscopes predominantly only enable imaging, with sufficient signal-to-noise ratio, in the upper layers of the mouse cortex (i.e. < 400 microns). Hence, to enable recordings throughout the depth of cortex, it is necessary to have a microscope with sufficiently high optical quality to enable high transmittance of laser power through the depth of the tissue. Additionally, it greatly simplifies user training and improves performance to have this microscope use the same control software as the acousto-optic scope (and hence to purchase both microscopes from the same manufacturer). Further, the data analysis pipeline can be duplicated across both microscopes, thereby decreasing the learning curve, and increasing scientific productivity.
The proposed pair of microscopes from Femtonics Ltd is the only available system which meets all the necessary criteria of the Department.

Clemson University's Department of Chemistry seeks to sole source the purchase of a Bruker 500MHz solid state nuclear magnetic resonance (NMR) spectrometer with multiphase and multinuclear capabilities. This instrument is essential for research outlined in a recently funded National Science Foundation Major Research Instrumentation (NSF-MRI) proposal. The NMR spectrometer comprises three main components: a superconducting magnet, probes, and a console that houses the electronics. Specifically, we require: 1. A high-resolution magic angle spinning (HRMAS) probe capable of 1H, 19F and BB acquisition. 2. A magic angle spinning (MAS) multinuclear probe capable of operating at temperatures from -80°C to 200°C. 3. Automatic magic angle adjustment and automatic shimming features, as the spectrometer will be used by non-experts. It is believed that Bruker Biospin Corporation is the only provider that offers a spectrometer and MAS probes meeting all of these specifications, including the Avance NEO NMR console, Bruker HRMAS iprobe, and CPMAS iprobe. No other supplier manufactures NMR instruments with these precise features. Furthermore, the Clemson NMR Facility currently operates three other Bruker NMR spectrometers. The high-resolution and MAS probes from these existing spectrometers are compatible with the proposed Bruker instrument and can serve as backup probes when the new ones are being serviced. This compatibility is specific to Bruker spectrometers. Additionally, the requested Bruker spectrometer includes the Ascend Evo superconducting magnet, which boasts a minimum helium hold time exceeding 365 days. Given the current global helium shortage, minimizing liquid helium consumption is crucial. No other manufacturer offers magnets with such low helium consumption. This equipment cannot be purchased from any other reseller or distributor. For these reasons, this is a sole source procurement.

Days are calculated as provided in §11-35-310(13). Both protests and notices of intent to protest must be in writing and must be received by the appropriate Chief Procurement Officer within the time provided. The grounds of the protest and the relief requested must be set forth with enough particularity to give notice of the issues to be decided.
